Given my views on play, you won’t be surprised to learn that I asked the presenters about whether play experiences could qualify as the kinds of responsive, positive, engaged, and empowering interactions they described. I was attending professional development aimed at helping library staff support families from refugee backgrounds, and we were discussing the role that libraries and childcare staff (mostly the latter, based on the mix of the group) can play in helping often highly traumatised communities. A small increase in risk of harm, replicated many times on a massive scale, means harm is certainly occurring. As COVID-19 lockdowns unfolded around the world, and sales of games, puzzles, toys, and other playful artforms spiked, a friend asked me if I felt a sense of validation. And worst of all, a combination of the two, where other players essentially become supporting cast for a scripted victory that is neither earned, nor fun, nor remotely playful – a psychodrama rather than a game, toying with their victims rather than playing with them. Controlling bullies who insist everyone play their way, forcing people to make deals they don’t want and to keep playing even when they aren’t having fun. Furthermore, miR-71 plays a prominent role in developmental recovery from L1 diapause partly through repressing the expression of certain heterochronic genes. Here we show that compromising overall microRNA (miRNA) functions or mutating certain individual miRNAs impairs the long-term survival of nematodes during starvation-induced L1 diapause. But offering people opportunities to explore games and play in a safe, non-abusive, self-directed context like a library seems to me to matter, and for folks who are more likely than most to have social anxieties, finding those opportunities can be extremely hard. This certainly accords with the rise of therapists using games as therapy – a web search for “Dungeons & Dragons therapy” shows the recent explosion in ways in which people are finding healing in play. That means that when sales are up, more people are playing this and other older, less-fun games instead of games with the advantage of more than a century’s advancements in the art and science of play, including co-operative games, social games, nomic games, and more. We identified a conserved “pause and play” mechanism of cell cycle during environmental stress and recovery. On 31 October, the band announced the 2025–2026 Tanzneid World Tour and released the song "Tanzneid" with a music video featuring Zummo on drums for the first time. This feels like one of those posts where if I had more time I could make it shorter… but I don’t. Fun seems self-evident, but I would just gently remind readers that fun is non-trivial; is in fact the active face of joy, as beauty is the passive. As above, play without consent, treating others as toys without independent will rather than as fellow players, is oppression and abuse. In fact, one of the great, almost pre-verbal lessons of play is precisely that the magic circle of play exists only with, is in large part comprised of, the willing participation of everyone in and around it. That curation isn’t only about having collections available for loan or in the library space; it’s also about making actual play experiences available.

To determine the functional relationship of miR-71 with LIN-42 and LIT-1, mir-71(lf); lin-42(lf) L1 worms were starved for 4 d and recovered on lit-1(RNAi) plates. The strong suppression of the mir-71(lf) defect by hbl-1(RNAi), and the relatively weak effect of miR-71 on hbl-1 expression, are consistent with the idea that miR-71 exerts its role by modulating activities of multiple genes related to hbl-1 function in developmental timing. We then compared the expression of a hbl-1 3′UTR reporter (18) in the mir-71(lf) mutants with that in wild type and found that the expression of this reporter was slightly derepressed at L3 in the mir-71 mutant (Fig. 4 F and G). We further examined worms recovering from 4 d of L1 starvation and found that around 90% of the mir-71(lf) mutants displayed retarded vulval precursor cell (VPC) division, compared with less than 5% in wild type (Fig. 4A). In contrast, the nuclear-localized GFP expression under the control of the 3′UTR of age-1(Fig. 3 C and D) or unc-31 (Fig. 3 E and F) was strongly repressed in the control worms, but prominently derepressed in mir-71(lf) mutant worms. If the 3′UTR of age-1 or unc-31 is repressed by miR-71, the GFP expression will be repressed in tissues where miR-71 is expressed in wild-type worms, but derepressed in the same tissues of mir-71(lf) worms. Among short-lived miRNA mutants, a mir-71 deletion mutant, mir-71(n4115) (referred to as mir-71(lf) hereafter), displayed a severe reduction in L1 starvation survival rate (Table S1 and Fig. 2A). Writing it out, playing both tapes forward, as plain as day on paper, did wonderful things. One who loves spending time with their family during the holidays and cooking delicious meals, despite the stress it sometimes brings on. The space for me to, for the first time in my adult life, truly decide how I want to live.

Components of the InsR pathway, including age-1, have recently been predicted to be targets of miR-71 in its role in aging (14). On the other hand, the role of a particular miRNA (miR-71) is executed by repressing the expression of many genes in multiple pathways. These results compelled us to examine specific interactions between individual miRNAs and their targets to gain mechanistic insights. This result is also consistent with the prediction from a miRISC immunoprecipitation analysis that hbl-1 is likely a target of one or more miRNAs, in addition to the let-7 family miRNAs, during early development (18). This result suggests that miR-71 likely functions upstream of, or in parallel to, HBL-1 in regulating VPC timing.

In a natural environment, periods of stress may be followed by recovery periods, when plants have the opportunity to return to normal growth conditions. Across the tree of life, organisms interact with the surrounding environment during growth and development. On 15 April, they announced their next album Tekkno, to be released on 9 September 2022. On 6 December, the band announced that they had applied to represent Germany in the 2022 Eurovision Song Contest with "Pump It", but ultimately were not included in the final list of participants. The coordinated entrance into developmental arrest, long-term survival, and the reinitiation of development upon food availability are important biological processes to investigate. Different organisms have developed versatile growth arrest strategies to overcome starvation-induced metabolic and developmental problems. Food deprivation is a life-threatening challenge that animals frequently face as individuals and as species. Numerous animal species across multiple phyla enter developmental arrest for long-term survival in unfavorable environments and resume development upon stress removal. By working toward these goals in a group setting, clients are able to track progress while receiving therapeutic input from trained clinicians. These sessions are not peer-led support groups, but professionally facilitated therapeutic spaces grounded in trauma-informed care. The overall effect of miRNAs on L1 starvation survival is expected to be significantly stronger than that reflected by the data in Fig. These results suggest that miRNAs act in the intestine, and possibly in other tissues, to promote L1 starvation survival. However, revery play the mechanisms that coordinate the long-term survival, overall developmental arrest, and reinitiation remain to be investigated.
